The Whore of Babylon was ancient Jerusalem. Prostitution in the bible was a metaphor for spiritual adultery, which means to be unfaithful to God by breaking covenant with Him. Jerusalem was the only nation in Covenant with God.

But the prophets say all nations streamed to Jeruslaem, was exalted above all other nations, and was the center of the earth. Jerusalem spiritually ruled the world. God sees things in through spirit not the flesh, Rome was an earthly power, but Israel had the Law and the Priesthood so in God's eyes Israel and Jerusalem ruled the world.
